Functional Assays

LysA™ Universal PARylation Assay Kit is a sandwich ELISA-based kit designed to measure and quantify the amount of total poly ADP-ribosylation present in cell extracts. The kit comes in a convenient 96-well format and contains all the reagents needed, including a PAR standard that allows to establish a standard curve for quantitative measurements. It contains enough antibodies, blocking buffer, and detection reagents to measure PARylation levels in cell extracts, in a 96 well plate format. It also includes cell lysates as controls for assay performance. Note: The assay is adequate for Poly-ADP-ribosylation detection only. The assay is linear in the 100 pM to 20 nM PARylation range. An anti-PAR antibody is used to coat the 96-well plate. Lysates from cells are added to the coated wells, so PAR (PARylated proteins) present in the cell lysates are captured by the antibody. The assay kit comes in a convenient 96-well format, with enough purified recombinant MAPKAPK2 kinase, kinase substrate, ATP, and kinase assay buffer for 100 enzyme reactions.

The NEU2 Fluorogenic Assay Kit is designed to measure NEU2 (Neuroaminidase 2 or Sialidase-2) glycohydrolase activity for screening and profiling applications. It comes in a convenient 96-well format, with purified recombinant NEU2 enzyme, NEU2 substrate, NEU2 assay buffer, and Stop Solution for 100 enzyme reactions. The NEU2 inhibitor NCGC00063279 is also included as a control. Hydrolysation of the NEU substrate releases a fluorophore (λexc/λem = 365 nm/445 nm). The increase in fluorescence at λ=445 nm is directly proportional to NEU2 activity. 

The STK3 (MST2) Kinase Assay Kit is designed to measure STK3 (serine/threonine-protein kinase 3) kinase activity for screening and profiling applications using Kinase-Glo® MAX as a detection reagent. The assay kit comes in a convenient 96-well format, with enough purified recombinant STK3 (MST2) kinase, kinase substrate, ATP, and kinase assay buffer for 100 enzyme reactions.

The Cathepsin D Inhibitor Screening Assay Kit is designed to measure the protease activity of Cathepsin D for screening and profiling applications. The Cathepsin D assay kit comes in a convenient 96-well or 384-well format, with enough recombinant human Cathepsin D (amino acids 21-412), its substrate, and Cathepsin buffer for 96 or 400 reactions. This kit contains Pepstatin A as internal control. The substrate is an internally quenched fluorogenic substrate. Proteolysis releases the highly fluorescent Mca from the quencher. Fluorescence intensity increases proportionally to the activity of the protease.
