Functional Assays

The NEU2 Fluorogenic Assay Kit is designed to measure NEU2 (Neuroaminidase 2 or Sialidase-2) glycohydrolase activity for screening and profiling applications. It comes in a convenient 96-well format, with purified recombinant NEU2 enzyme, NEU2 substrate, NEU2 assay buffer, and Stop Solution for 100 enzyme reactions. The NEU2 inhibitor NCGC00063279 is also included as a control. Hydrolysation of the NEU substrate releases a fluorophore (λexc/λem = 365 nm/445 nm). The increase in fluorescence at λ=445 nm is directly proportional to NEU2 activity. 

The STK3 (MST2) Kinase Assay Kit is designed to measure STK3 (serine/threonine-protein kinase 3) kinase activity for screening and profiling applications using Kinase-Glo® MAX as a detection reagent. The assay kit comes in a convenient 96-well format, with enough purified recombinant STK3 (MST2) kinase, kinase substrate, ATP, and kinase assay buffer for 100 enzyme reactions.

The Cathepsin D Inhibitor Screening Assay Kit is designed to measure the protease activity of Cathepsin D for screening and profiling applications. The Cathepsin D assay kit comes in a convenient 96-well or 384-well format, with enough recombinant human Cathepsin D (amino acids 21-412), its substrate, and Cathepsin buffer for 96 or 400 reactions. This kit contains Pepstatin A as internal control. The substrate is an internally quenched fluorogenic substrate. Proteolysis releases the highly fluorescent Mca from the quencher. Fluorescence intensity increases proportionally to the activity of the protease.

The Sialidase Fluorogenic Assay Kit is designed to measure Sialidase NANH (N-acylneuraminate glycohydrolase) activity for screening and profiling applications. It comes in a convenient 96-well format, with purified recombinant Salmonella typhimurium sialidase, Sialidase substrate, Sialidase assay buffer, and Stop Solution for 100 enzyme reactions. Sialidase inhibitor NCGC00063279 (N-Acetyl-2,3-dehydro-2-deoxyneuraminic acid) is also included as a control. The Sialidase fluorogenic substrate is incubated with Sialidase. Hydrolyzation of the Sialidase substrate releases a fluorophore (λexc/λem = 365 nm/445 nm). The increase in fluorescence at λ=445 nm is directly proportional to Sialidase activity. 

The ARH3 Fluorogenic Assay Kit is a high-throughput, homogeneous 96-well assay designed to measure the hydrolase activity of ARH3 (ADP-ribosylhydrolase) for screening and profiling applications, using a simple and straightforward fluorogenic assay. The ARH3 Fluorogenic Assay Kit contains enough purified recombinant ARH3 enzyme, substrate, and assay buffer for 100 enzyme reactions. The kit includes the ARH3 inhibitor ADP-HPD as an ARH3 activity control. ARH3 is incubated with a fluorogenic ADP-ribose substrate in which the fluorophore is quenched by the presence of the ribose. ARH3-mediated hydrolysis of the substrate between the ribose and the fluorochrome releases fluorescence that can be detected at λ=502 nm (excitation at λ=385 nm).
