Functional Assays

The Chemi-Verse™ PI3 Kinase (p110α/p85α) Kinase Assay Kit is designed to measure kinase activity of the PI3 kinase (p110α/p85α) complex for screening and profiling applications using ADP-Glo™ as a detection reagent. The assay kit comes in a convenient 96-well format, with enough purified recombinant PI3 kinase (p110α/p85α), kinase substrate, ATP, and kinase assay buffer for 100 enzyme reactions.

The Chemi-Verse™ PI3 Kinase p110α (E545K)/p85α Kinase Assay Kit is designed to measure the kinase activity of the complex PI3 (phosphoinositide 3) kinase consisting of mutant p110α (E545K) and p85α for screening and profiling applications, using ADP-Glo™ as a detection reagent. The assay kit comes in a convenient 96-well format, with enough purified recombinant human complex PI3 Kinase p110α (E545K)/p85α kinase, kinase substrate, ATP, and kinase assay buffer for 100 enzyme reactions. 

The Cathepsin E Inhibitor Screening Assay Kit is designed to measure the protease activity of Cathepsin E for screening and profiling applications. The Cathepsin E assay kit comes in a convenient 96-well or 384-well format, with enough recombinant human Cathepsin E (amino acids 18-396), its substrate, and Cathepsin buffer for 96 or 384 reactions. This kit includes inhibitor Pepstatin A as control. The substrate is an internally quenched fluorogenic substrate. Proteolysis releases the highly fluorescent Mca from the quencher. Fluorescence intensity increases proportionally to the activity of the protease.

This assay uses o-phthalaldehyde and DTT (dithiothreitol) as detection reagents. The assay kit comes in a convenient 96-well format, with enough purified recombinant PADI4, BAEE (Nα-Benzoyl-L-arginine ethyl ester hydrochloride), EDTA (Ethylenediaminetetraacetic acid), o-phthalaldehyde, and assay buffer for 100 reactions.

LysA™ Universal PARylation Assay Kit is a sandwich ELISA-based kit designed to measure and quantify the amount of total poly ADP-ribosylation present in cell extracts. The kit comes in a convenient 96-well format and contains all the reagents needed, including a PAR standard that allows to establish a standard curve for quantitative measurements. It contains enough antibodies, blocking buffer, and detection reagents to measure PARylation levels in cell extracts, in a 96 well plate format. It also includes cell lysates as controls for assay performance. Note: The assay is adequate for Poly-ADP-ribosylation detection only. The assay is linear in the 100 pM to 20 nM PARylation range. An anti-PAR antibody is used to coat the 96-well plate. Lysates from cells are added to the coated wells, so PAR (PARylated proteins) present in the cell lysates are captured by the antibody.
