Cryopreserved Human Peripheral Blood Mononuclear Cells (PBMCs) are available as positive and negative controls for T-cell monitoring in ELISPOT, ELISA, cytokine bead array, tetramer/pentamer, and flow cytometry assays. A peripheral blood mononuclear cell (PBMC) is defined as any blood cell with a round nucleus (i.e. a lymphocyte, a monocyte, or a macrophage). These blood cells are a critical component in the immune system to fight infection and adapt to intruders. The lymphocyte population consists of T cells, B cells and Natural Killer cells. These cells are often extracted from whole blood or from leukopacks using ficoll, a hydrophilic polysaccharide that separates layers of blood, with monocytes and lymphocytes forming a buffy coat under a layer of plasma. Samples from each donor are tested via PCR to confirm non-reactivity.

The organization of the epithelium of the small intestine and colon into crypts is generally comparable. However, histologically there are two important differences between the two types of epithelia.The colonic epithelium has a flat epithelium surface and does not have villi. Regeneration occurs from units embedded within the crypts, each containing about 6–10 stem cells, while differentiated epithelial cells reside at the surface. Human colonic epithelium is linked to various human diseases and disorders such as Ulcerative Colitis and Colon Cancer, necessitating a better understanding of epithelial cell function and their role in disease-affected tissues. CAI’s colonic epithelial culture system provides a unique tool for the investigation of colon epithelial cell physiology and evaluation of therapeutic mediators for translational research.

We offer pooled canine hepatic subcellular fractions isolated from Beagle dog liver tissue, demonstrating high enzyme activity level. Options include matching microsomal, S9, and cytosolic fractions from male or female donor pools to minimize variation in canine liver enzyme levels. We also offer dog liver microsomes treated with prototypical inducers for use as a positive control in ex vivo enzyme induction or other drug metabolism studies. Microsomes are supplied in 0.5 mL volume at a concentration of 20 mg/mL in 250 mM sucrose buffer. S9 (1 mL at 20 mg/mL) and cytosol (1 mL at 10 mg/mL) fractions are packaged in a suspension buffer containing 50 mM TRIS-HCI, (pH 7.4 at 4ºC), 150 mM KCI, and 2 mM EDTA.
