Many small fluorescent probes are used to monitor lysosomes in living cells. Dojindo’s LysoPrime Deep Red overcomes known problems with fluorescent lysosome probes, such as lack of specificity for lysosomes and staining dependent on the lysosomal pH. In addition, the high-retentivity of LysoPrime Deep Red enables long-term imaging experiments.

Live imaging using small molecule fluorescent probes has been widely used for lysosomal live cell analysis, but the low specificity and retention ability due to pH change have been cited as issues. The pHLys Red is a small molecule fluorescent probe with high lysosomal specificity and sensitivity to pH changes, enabling a more accurate analysis of lysosomal pH in live cells. It is also applicable to experiments that require long-term imaging due to its high retention ability.

This kit includes lysosome staining dyes, pHLys Red (pH dependent), and LysoPrime Green (pH-independent). The pHLys Red and LysoPrime Green accumulate in the intact lysosomes. The fluorescence intensity of pHLys Red is enhanced as the acidity increases, and weak fluorescence is observed when lysosomes are neutralized due to the lysosomal dysfunction. On the other hand, LysoPrime Green gives stable emissions even lysosomes are neutralized. Lysosomal pH and lysosomal mass can be measured by combining these two dyes.
