By stimulating PBMCs with infection-specific peptides, a targeted T cell response is initiated and is visualized directly on the plate surface. As T cells secrete cytokines in response to stimulation, the cytokines are captured by antibodies around the cell on the surface of the well and visualised via a colorimetric reaction. Each cytokine-secreting cell forms a distinct "spot", creating a visual footprint that provides a precise count of responding cells.
