Caspase 6 is a cysteine protease involved in the execution phase of apoptosis mediating neuronal shrinkage. In addition it has important roles in neurodegenerative disorders such as Huntington’s Parkinson’s and Alzheimer’s diseases. Caspase 6 cleaves the mutated huntintin protein and the amyloid precursor protein resulting in the aggregating protein fragments typical of Huntington’s and Alzheimer’s diseases respectively.Activity is quantified by release of free fluorescent 7-amino-4-trifluoromethylcoumarin (AFC) which excites at 395-400 nm and emits at 495-505 nm.References1.
