1-Naphthalenesulfonyl-Ile-Trp-aldehyde is a a reversible inhibitor of Cathepsin L (IC₅₀ = 1.9 nM). Cathepsin L is a lysosomal cysteine protease with an important role in initiating protein degradation. Cathepsin L is also upregulated in several cancers and has been suggested as a drug target. In addition, extracellular secretion of Cathepsin L by tumors implicates it as a potential serum biomarker for cancer progression. In an in vitro bone culture system it inhibits the release of hydroxyproline and Ca²⁺ and prevents bone loss in ovariectomized mice (~50 mg/kg).
