Cas9 (CRISPR-associated protein 9) is a 160 kD enzyme that plays an important role in bacterial defense against DNA viruses and plasmids, and is heavily used in CRISPR genome editing. Cas9 has a two-lobed structure, with a single bridge between the alpha-helical lobe and the nuclease lobe. In CRISPR, a guide RNA binds to the alpha-helical lobe and a specific region of DNA, then the nuclease lobe cleaves both strands of the DNA. CRISPR with Cas9 enables scientists to edit genomic DNA via addition, deletion or substitution. Cas9 is comprised of 1368 amino acids and the GenPept accession number is WP_038431314. The HA (human influenza hemagglutinin) tag is a 9-amino acid peptide commonly used to label proteins in biomedical research, and can be detected with anti-HA antibodies.
