Her2 (hormone epidermal growth factor receptor 2) is a type I membrane receptor kinase encoded by the ERBB2 gene and overexpressed in a variety of cancers including breast, gastric, lung and ovarian cancers. Her2 expression is associated with the development and progression of certain aggressive types of breast cancer. ProMab’s PM-LNP-0018 nanoparticles contain an mRNA encoding an anti-Her2 CAR (chimeric antigen receptor) protected by a lipid shell. The CAR contains a signal sequence, the antiHer2 scFv (single-chain variable fragment), the CD8 transmembrane domain, the 4-1BB costimulatory domain, and the CD3ζ activation domain. The nanoparticles are formulated with SM-102, DSPC, cholesterol and DMGPEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery in vitro and in vivo.
