TMB ELISA Peroxidase Substrate (3, 3', 5, 5' - Tetramethylbenzidine) is a chromogenic substrate used to visualize antibody reactivity in ELISA experiments. In the presence of peroxidases, TMB can act as an electron donor for the conversion of peroxides to water, typically changing the color of solution to a blue color equivalent to the degree of reactivity and upon addition of acid, changing the solution color to yellow. This proprietary formulation of TMB ELISA Peroxidase Substrate is red in color, enabling users to have visual certainty that the product was applied equally. TMB ELISA Peroxidase Substrate is ideal for investigators in Immunology, Cancer, and Microbiology research.

    • TMB Membrane Peroxidase Substrate (3, 3', 5, 5' - Tetramethylbenzidine) is a chromogenic substrate used to visualize antibody reactivity in ELISA, WB, or IHC experiments. In the presence of peroxidases, TMB can act as an electron donor for the conversion of peroxides to water, changing the color of solution to a blue color equivalent to the degree of reactivity. This reaction of TMB can be halted with acid to change the TMB to yellow. TMB Membrane Peroxidase Substrate is ideal for investigators in Microbiology, Immunology, and Cancer research.

    • This following method is suggested as a guideline for the use for Western Blot with nitrocellulose or PVDF membranes. The TMB Substrate in 100 mL bottle TMBM-101 and the TMB Precipitating Agent in 10 mL bottle TMBM-102 are used together. Mix 9 ml of the TMB Substrate and 1.0 ml of the TMB Precipitating Agent and mix thoroughly. The substrate is now ready to use. Be certain to use within 24 hours. After antibody incubations and wash, aspirate the wash solution and add approximately 5 ml of freshly prepared substrate solution. Incubate until color develops (usually 2 to 20 min). Wash the membrane with 2 changes of deionized water for 5 min each.

    • The one component 3,3?,5,5?-tetramethylbenzidine (TMB) substrate is supplied in a ready-to-use solution. Peroxidase reacts with the TMB substrate solution to produce a soluble blue reaction product that can be read at 370 nm or 620-650 nm for ELISA applications. Stop solutions can be added to prevent further color development and reading the absorbance values at the desired wavelengths. Benefits of the BioFX family of substrates include: Prolonged stability. Increased sensitivity. Neglibible backgrounds. Ease of Manufacturing.

    • TMB Precipitating (K-TMBP) is a chromogen substrate that reacts with horseradish peroxide (HRP) producing a permanent purple colored spot or band. This detection reagent is best used with an HRP conjugated secondary or tertiary antibody on a dot blot or western blot. We recommend K-TMBP for use with lipid strips and arrays.
